    3D model of amphioxus steroid receptor complexed with estradiol

    The origins of signaling by vertebrate steroids are not fully understood. An important advance was the report that an estrogen-binding steroid receptor [SR] is present in amphioxus, a basal chordate with a similar body plan as vertebrates. To investigate the evolution of estrogen binding to steroid receptors, we constructed a 3D model of amphioxus SR complexed with estradiol. This 3D model indicates that although the SR is activated by estradiol, some interactions between estradiol and human ER[alpha] are not conserved in the SR, which can explain the low affinity of estradiol for the SR. These differences between the SR and ER[alpha] in the steroid-binding domain are sufficient to suggest that another steroid is the physiological regulator of the SR. The 3D model predicts that mutation of Glu-346 to Gln will increase the affinity of testosterone for amphioxus SR and elucidate the evolution of steroid binding to nuclear receptors

    Assessing housing quality and its impact on health, safety and sustainability

    Background The adverse health and environmental effects of poor housing quality are well established. A central requirement for evidence-based policies and programmes to improve housing standards is a valid, reliable and practical way of measuring housing quality that is supported by policy agencies, the housing sector, researchers and the public. Methods This paper provides guidance on the development of housing quality-assessment tools that link practical measures of housing conditions to their effects on health, safety and sustainability, with particular reference to tools developed in New Zealand and England. Results The authors describe how information on housing quality can support individuals, agencies and the private sector to make worthwhile improvements to the health, safety and sustainability of housing. The information gathered and the resultant tools developed should be guided by the multiple purposes and end users of this information. Other important issues outlined include deciding on the scope, detailed content, practical administration issues and how the information will be analysed and summarised for its intended end users. There are likely to be considerable benefits from increased international collaboration and standardisation of approaches to measuring housing hazards. At the same time, these assessment approaches need to consider local factors such as climate, geography, culture, predominating building practices, important housing-related health issues and existing building codes. Conclusions An effective housing quality-assessment tool has a central role in supporting improvements to housing. The issues discussed in this paper are designed to motivate and assist the development of such tools

    Explosive Event in MON-3 Oxidizer System Resulting from Pressure Transducer Failure

    In 2003, a Druck(Registered Trademark) pressure transducer failed catastrophically in a test system circulating nitrogen tetroxide at NASA Johnson Space Center White Sands Test Facility. The cause of the explosion was not immediately obvious since the wetted areas of the pressure transducer were constructed of materials compatible with nitrogen tetroxide. Chemical analysis of the resulting residue and a materials analysis of the diaphragm and its weld zones were used to determine the chain of events that led to the catastrophic failure. Due to excessive dynamic pressure loading in the test system, the diaphragm in the pressure transducer suffered cyclic failure and allowed the silicon oil located behind the isolation diaphragm to mix with the nitrogen tetroxide. The reaction between these two chemicals formed a combination of 2,4-di and 2,4,6-trinitrophenol, which are shock sensitive explosives that caused the failure of the pressure transducer. Further research indicated numerous manufacturers offer similar pressure transducers with silicone oil separated from the test fluid by a thin stainless steel isolation diaphragm. Caution must be exercised when purchasing a pressure transducer for a particular system to avoid costly failures and test system contamination

    Lattice Monte Carlo calculations for unitary fermions in a finite box

    We perform lattice Monte Carlo simulations for up to 66 unitary fermions in a finite box using a highly improved lattice action for nonrelativistic spin 1/2 fermions. We obtain a value of 0.3660.011+0.0160.366^{+0.016}_{-0.011} for the Bertsch parameter, defined as the energy of the unitary Fermi gas measured in units of the free gas energy in the thermodynamic limit. In addition, for up to four unitary fermions, we compute the spectrum of the lattice theory by exact diagonalization of the transfer matrix projected onto irreducible representations of the octahedral group for small to moderate size lattices, providing an independent check of our few-body simulation results. We compare our exact numerical and simulation results for the spectrum to benchmark studies of other research groups, as well as perform an extended analysis of our lattice action improvement scheme, including an analysis of the errors associated with higher partial waves and finite temporal discretization.Comment: Significant revisions from previous version.     Eurovision 2023 Cultural Relations Snapshot: A snapshot from the forthcoming cultural relations, soft power and shared values research

    About this research In May 2023, Liverpool and the BBC hosted the Eurovision Song Contest on Ukraine’s behalf. This was the first time since 1980 that Eurovision has not been hosted in the previous winning country, and the first time a winner has ever been unable to host because the country is under attack. Researchers at the University of Hull with consultants from the University of Brighton, University of Glasgow, and Royal Holloway, University of London are investigating the cultural relations and ‘soft power’ impact of Eurovision for a study commissioned by the British Council in partnership with Liverpool City Council and DCMS. The study has three pillars: • A literature review surveying the state of the art in research on Eurovision and other largescale cultural and competitive events. • A tracker survey exploring the impact of Eurovision on members of the public in 5 European countries. • Interviews exploring how stakeholders co-operated to create the narrative of Liverpool, the BBC and the UK hosting Eurovision on behalf of Ukraine. We’ve prepared this snapshot ahead of the publication of the full research in January 2023 for the upcoming event ‘Liverpool Calling – And the results are in!’

    Intra-session and inter-day reliability of the Myon 320 electromyography system during sub-maximal contractions

    Electromyography systems are widely used within the field of scientific and clinical practices. The reliability of these systems are paramount when conducting research. The reliability of Myon 320 Surface Electromyography System is yet to be determined. This study aims to determine the intra-session and inter-day reliability of the Myon 320 Surface Electromyography System. Muscle activity from fifteen participants was measured at the anterior deltoid muscle during a bilateral front raise exercise, the vastus lateralis muscle during a squat exercise and the extensor carpi radialis brevis (ECRB) muscle during an isometric handgrip task. Intra-session and inter-day reliability was calculated by intraclass correlation coefficient, standard error of measurement and coefficient of variation (CV). The normalized root mean squared (RMS) surface electromyographic signals produced good intra-session and inter-day testing intraclass correlation coefficient values (range: 0.63-0.97) together with low standard error of measurement (range: 1.49-2.32) and CV (range: 95% Confidence Interval = 0.36-12.71) measures for the dynamic-and-isometric contractions. The findings indicate that the Myon 320 Surface Electromyography System produces good to fair reliability when examining intra-session and inter-day reliability. Findings of the study provide evidence of the reliability of electromyography between trials which is essential during clinical testing.</p

    Simultaneous multiple allelic replacement in the malaria parasite enables dissection of PKG function.

    Over recent years, a plethora of new genetic tools has transformed conditional engineering of the malaria parasite genome, allowing functional dissection of essential genes in the asexual and sexual blood stages that cause pathology or are required for disease transmission, respectively. Important challenges remain, including the desirability to complement conditional mutants with a correctly regulated second gene copy to confirm that observed phenotypes are due solely to loss of gene function and to analyse structure-function relationships. To meet this challenge, here we combine the dimerisable Cre (DiCre) system with the use of multiple lox sites to simultaneously generate multiple recombination events of the same gene. We focused on the Plasmodium falciparum cGMP-dependent protein kinase (PKG), creating in parallel conditional disruption of the gene plus up to two allelic replacements. We use the approach to demonstrate that PKG has no scaffolding or adaptor role in intraerythrocytic development, acting solely at merozoite egress. We also show that a phosphorylation-deficient PKG is functionally incompetent. Our method provides valuable new tools for analysis of gene function in the malaria parasite
